چکیده

Inositol 1 ,4,5-trisphosphate (InsP3) 3-kinase is responsible for the synthesis of InsP4. The kinase in many tissues is regulated by Ca2+/calmodulin (CaM; see [1] for review). The activity of purified enzyme from bovine brain is stimulated up to 30-fold by Ca2+/CaM. Stimulation by CaM appears to result from an increase in enzyme maximal velocity without a substantial change of Km for InsP3 [2]. Similar changes in kinetics have been reported for another well-characterized CaM-regulated enzyme, the cyclic nucleotide CaM-sensitive phosphodiesterase [3]. Increases of activity of that enzyme are also 20-40-fold when assayed in the presence of Ca2` and saturated CaM

Long lasting inhibition of adenylyl cyclase selectively mediated by inositol 1,4,5-trisphosphate-evoked calcium release.

In A7r5 smooth muscle cells, vasopressin stimulates release of Ca2+ from intracellular stores and Ca2+ entry, and it inhibits adenylyl cyclase (AC) activity. Inhibition of AC is prevented by inhibition of phospholipase C or when the increase in cytosolic [Ca2+] is prevented by the Ca2+ buffer, BAPTA. It is unaffected by pertussis toxin, inhibition of protein kinase C, or L-type Ca2+ channels or...

Cloning and expression in Escherichia coli of a rat brain cDNA encoding a Ca2+/calmodulin-sensitive inositol 1,4,5-trisphosphate 3-kinase.

Inositol 1,4,5-trisphosphate (InsP3) 3-kinase catalyses the phosphorylation of InsP3 to inositol 1,3,4,5-tetrakisphosphate (InsP4). InsP3 3-kinase activity was stimulated by Ca2+ in the presence of calmodulin (CaM) and the protein was associated with two silver-stained bands which migrated with an apparent Mr of approx. 50,000 on SDS/polyacrylamide gels. Upon limited proteolysis with trypsin, t...

The role of calmodulin-binding sites in the regulation of the Drosophila TRPL cation channel expressed in Xenopus laevis oocytes by ca2+, inositol 1,4,5-trisphosphate and GTP-binding proteins.

The roles of calmodulin-binding sites in the regulation by Ca2+, inositol 1,4,5-trisphosphate (InsP3) and GTP-binding regulatory proteins (G-proteins) of the Drosophila melanogaster TRPL (transient-receptor-potential-like) non-specific Ca2+ channel were investigated. Wild-type TRPL protein and two mutant forms, TRPL (W713G) and TRPL (W814G), in which a key tryptophan residue in each of the two ...

Norepinephrine-induced inositol 1,4,5-trisphosphate formation in atrial myocytes is regulated by extracellular calcium, protein kinase C, and calmodulin.

We investigated whether alteration of extracellular and intracellular Ca2+ concentrations, protein kinase C, and calmodulin modulate norepinephrine (NE)-induced inositol 1,4,5-trisphosphate (IP3) formation in neonatal rat atrial myocytes. NE-induced IP3 production in atrial myocytes was stimulated by elevation of extracellular Ca2+ in a dose-dependent manner. However, TMB-8 (an intracellular ca...

Cation sensitivity of inositol 1,4,5-trisphosphate production and metabolism in agonist-stimulated adrenal glomerulosa cells.

Angiotensin II (AII) evokes a biphasic increase in inositol 1,4,5-trisphosphate (Ins(1,4,5)P3) levels in adrenal glomerulosa cells, with an extracellular Ca(2+)-independent early peak followed by a secondary sustained elevation that is highly dependent on the presence of extracellular Ca2+. The Ca(2+)-dependent sustained phase of agonist-induced Ins(1,4,5)P3 production was closely correlated wi...
